摘要
目的:探索CCAAT/增强子结合蛋白α(C/EBPα)基因对HeLa细胞增殖和定向迁移的影响。方法:用MTT方法检测C/EBPα基因重组质粒转染组、空质粒转染组和未转染组对HeLa细胞增殖的影响。用Transwell侵袭转移模型评价He-La细胞的迁移情况。其中,不同上室中分别加入C/EBPα基因重组质粒转染组、空质粒转染组和未转染组的HeLa细胞,下室中加入含20%FBS的培养基。结果:C/EBPα基因重组质粒转染组、转染空质粒组和未转染HeLa细胞组的吸光度值分别为0.21±0.01、0.24±0.01和0.24±0.02,重组质粒转染组与空质粒转染组和未转染HeLa细胞组比较差异有统计学意义(P<0.05)。Transwell实验发现重组质粒转染组与空质粒转染组和未转染HeLa细胞组相比,细胞迁移能力减弱,细胞穿膜数分别为12.0±2.2、39.0±3.2和39.6±2.6,重组质粒转染组与空质粒转染组和未转染HeLa细胞组比较差异有统计学意义(P<0.05)。结论:C/EBPα基因能抑制子宫颈癌HeLa细胞的增殖和迁移能力。
Objective: To explore the effect of CCAAT/enhancer binding protein α (C/EBPα) gene on proliferation and orient immigration of HeLa cells. Methods: MTr method was used to detect the effects of C/EBPα gene recombinant plasmid transfection group, empty plasmid group, and non - transfection group on proliferation of HeLa cells. Transwell invasion and metastasis model was used to evaluate migration of HeLa cells. HeLa cells in C/EBPα gene recombinant plasmid transfection group, empty plasmid group, and non - transfec- tion group were placed into different upper transwells, and culture medium containing 20% FBS were placed into the lower transwells, results: The values of optical density of HeLa cells in C/EBPα gene recombinant plasmid transfection group, empty plasmid group, and non - transfection group were ( 0. 21 ±0. 01 ), (0. 24 ± 0. 01 ), and (0. 24 ± 0.02 ), respectively. There was statistically significant difference between C/EBPα gene recombinant plasmid transfection group, empty plasmid group and non - transfection group (P 〈 0.05 ) . Transwell test found that compared with HeLa cells in non - transfection group, the migrant ability of HeLa cells weakened in C/EBPα gene recombinant plasmid transfection group and empty plasmid group, the numbers of transfected cells through membrane were ( 12. 0 ± 2. 2 ) , (39. 0± 3.2 ) , and (39. 6± 2. 6 ) , respectively, there was statistically significant difference between C/EBPα gene recombinant plasmid transfeetion group, empty plasmid group and non - transfection group ( P 〈 0. 05 ) . Conclusion: C/EBPα gene can inhibit the proliferative ability and migrant ability of cervical cancer HeLa cells.
