蛔虫体腔液对3T3-L1细胞分化和凋亡的作用

摘要 为了探讨蛔虫体腔液(Ascaris body cavity fluid,ABF)对小鼠3T3-L1前脂肪细胞分化过程的作用,揭示ABF对机体脂类代谢的影响。本试验采用半定量聚合酶链式反应技术(RT-PCR)检测前脂肪细胞分化相关基因过氧化物酶体增殖剂活化受体γ(Peroxisome proliferator-activated receptorγ,PPARγ)、GAPDH mRNA的表达;流式细胞技术(FCM)检测ABF对3T3-L1细胞凋亡的影响。结果表明:1 000μg/mL的ABF明显降低前脂肪细胞分化相关基因PPARγmRNA的相对表达,与对照相比,差异极显著(P<0.01)。在3T3-L1分化过程中加入1 000μg/mL ABF可增加细胞的凋亡。表明一定浓度的ABF对3T3-L1前脂肪细胞分化具有明显的抑制作用,并且能够诱导3T3-L1前脂肪细胞分化过程中的细胞凋亡,提示ABF能够抑制前脂肪细胞转化为脂肪细胞,可能具有调控脂类代谢以及控制高脂血症相关疾病的潜力。 To investigate the effects of Ascaris body cavity fluid(ABF) on differentiation and apoptosis of 3T3-L1 preadipocytes, the expressions of PPAR7 mRNA and GAPDH mRNA were measured by quantitative real time PCR. Apoptosis rates in the stage of differentiation in 3T3-L1 cells were detected by flow cytometric analysis before and after the treatment of Ascaris body cavity fluid. The results showed that 1 000μg/mL ABF significantly reduced mRNA expressions of PPART. The apoptosis rate was increased during 3T3-L1 differentiation by adding 1 000μg/mL ABF. 1 000 μg/mL ABF had a significant cytotoxic effect on 3T3-L1 prea- dipocytes, and significantly induced apoptosis. ABF inhibited the differentiation of preadipoeytes into adipocytes, and may have the potential of regulation of lipid metabolism and control of hyperlipidemia.
