摘要
目的研究逍遥散抗抑郁有效部位(A,B4,B3)配伍对应激抑郁模型小鼠行为学及睡眠时间的影响。方法逍遥散抗抑郁有效部位采用L9(34)正交设计进行配伍研究,以配伍后的组份及文拉法辛为干预药物,灌胃给予15 d的实验药物。实验过程中,定期测定小鼠体重与自主活动。于第14天给药后1 h,采用急性应激抑郁模型(小鼠悬尾)观测配伍组份对小鼠悬尾不动时间的影响,初步筛选最佳配伍比;第15天给药后1 h,各组小鼠按45 mg/kg腹腔注射阈剂量的戊巴比妥钠生理盐水注射液,准确记录注射结束时间、翻正反射消失时间、翻正反射恢复时间。观测配伍组份对戊巴比妥钠所致小鼠睡眠时间的影响。结果与空白对照组比较,当配伍部位按生药量计,A为138.9 g/kg,B4为46.3 g/kg,B3为23.1 g/kg时,可显著减少小鼠悬尾不动时间(P<0.001),且不影响小鼠自主活动量,并能够明显延长小鼠睡眠潜伏期(P<0.01)和显著地缩短小鼠睡眠持续时间(P<0.001)。结论逍遥散抗抑郁有效部位按生药量计,A为138.9 g/kg,B4为46.3 g/kg,B3为23.1 g/kg时,抗抑郁作用最佳,且在最佳配伍比时,能明显缩短阈剂量戊巴比妥钠所致小鼠睡眠时间。
Objective To study the effect of the compatibility of three active fractions from Xiaoyaosan on changes of behavior and sleep time on stress model in mice. Methods The active fractions(A,B4,B3) were combined into different compatibility groups using L9(34) orthogonal design.The mice were treated with the compatibility groups or positive drug(venlafaxine) for 15 days.During the experiment,body weight,data of open-field test(OFT) were detected.On the 14th day,the tail suspension test(TST) in mice was conducted 1 h after the drug treatment to screen the best anti-depression compatibility group.1 h after the last drug treatment,ip 45 mg/kg barbital sodium.To write down ip time,disappeance time of righting reflex,refreshment time of righting reflex. Results The locomotor activity in mice OFT was not affected by the treatment of drugs.The immobility time of the mice in the mouse TST treated with active fractions(A:138.9 g/kg,B4:46.3 g/kg,B3:23.1 g/kg) increased significantly(P〈0.001) compared with control group,the compatibility group extended mice sleep latency and decreased sleep time significantly(compare with control group,P〈0.01 and P〈0.001,respectively). Conclusion The compatibility group(A:138.9 g/kg,B4:46.3 g/kg,B3:23.1 g/kg) has the best anti-depression effect and can decrease sleep time.
