烟草光能兼养型愈伤组织的诱导和培养条件 被引量:3

摘要 以烟草SR1品种的4周龄叶片为外植体,使用MS+2 mg/L 2,4 D(2,4-二氯苯氧乙酸)+0.25mg/L KT(6-糖基氨基嘌呤)+3%蔗糖+0.8%琼脂的固体培养基成功诱导了叶片的愈伤组织.通过改变蔗糖浓度和激素配比,对兼性愈伤组织的培养基进行了筛选.结果表明:在500 lxs光照下,蔗糖浓度由3%降至1.5%时,愈伤组织有变绿趋势;蔗糖浓度<1.5%时,愈伤组织容易出现褐化.2 mg/L NAA(α-萘乙酸)+0.25 mg/L KT的激素组合可有效诱导愈伤组织的光合兼养,而单一植物激素如NAA和6-BA(6-苄氨基腺嘌呤),不是理想的诱导配方. In this study, 4 weeks of tobacco cultuivar-SR1 leaves were used as the explants and solid culuter medium containing MS + 2 mg/L 2,4-D ( 2,4-dichlorophenoxy ) + 0.25 mg/L KT (kinetin) + 3 % sucrose + 0.8 % agar was employed to successfully induce the callus formation. Then culture mediums aiming for photoautotrophic facultative callus culture were screened by altering the sucrose concentrations and hormone combinations. Under the irradiation of 500 lxs light intensity, when the concentration of sucrose was reduced from 3% to 1. 5%, the callus tissue was found to increasingly turn green. However, when the concentration of sucrose was reduced to below 1.5% , it easily turned brown. The combination of 2 mg/L NAA (1-naphthlcetic) with 0. 25 mg/L KT can effectively induce the production of photoautotrophic facultative callus, whereas the unique plant hormone, such as NAA or 6-BA (6-benzylaminopurine) can not be used as the ideal medium for such induction.
