摘要 卵巢癌是恶性程度很高的妇科肿瘤之一.在一些卵巢癌组织中常常存在p53基因的突变.p53基因突变和第72密码子多态性在癌症诊断中的作用目前尚不明确.有研究表明,p53肿瘤抑制基因Arg72Pro多态性与卵巢癌相关.然而,各研究结果不一致.本meta分析旨在探讨Arg72Pro多态性与卵巢癌之间的关系.在PubMed数据库中查询文献,包含Arg72Pro基因型频率的病例对照研究文献纳入本研究,并计算肿瘤发生危险性的优势比(odd ratio,OR)及其95%置信区间(confidence interval,CI).分析结果显示在各遗传模式下p53的Arg72Pro多态性与肿瘤发病风险的相关性均不显著(加性模式:OR=1.045,95%CI 0.953~1.146,P=0.354;显性模式:OR=1.096,95%CI 0.971~1.238,P=0.137;隐性模式:OR=0.962,95%CI 0.789~1.173,P=0.7).p53的Arg72Pro多态性与卵巢癌发病风险提高可能并不存在关联. Ovarian cancer is the leading cause of death from gynecological malignancies, p53 gene is fre- quently mutated in some ovarian carcinomas. The role of p53 mutations and polymorphic variant of codon 72 in the prognosis of disease is still unclear, p53 tumor suppressor gene Arg72Pro polymorphism have been as- sociated with ovarian cancer. However, results were inconsistent. This meta-analysis was performed to esti- mate the association between p53 Arg72Pro polymorphism and ovarian cancer. Electronic search of PubMed was conducted to select studies. Studies containing available genotype frequencies of Arg72Pro were chosen and pooled odds ratio (OR) with 95% CI ( confidence interval) was used to assess the association. The o- verall results suggested that the variant genotypes were not associated with the ovarian cancer risk in all ge- netic models (additive model: OR = 1. 045, 95% CI 0. 953 - 1. 146,P = 0. 354;dominant model: OR = 1. 096, 95% CIO. 971 - 1. 238,P = 0. 137 ; recessive model: OR =0. 962, 95% CIO. 789 - 1. 173,P = 0.7 ). This meta-analysis reveals that p53 Arg72Pro polymorphism is not associated with increased risk of ovarian cancer.
