摘要
目的:探索脂多糖(lipopolysaccharide,LPS)刺激下水通道蛋白5(aquaporin-5,AQP5)和黏蛋白5AC(mucin 5AC,MUC5AC)的变化。方法:采用不同的时间点、不同浓度LPS干预SPC-A1细胞株,荧光定量聚合酶链反应(polymerase chainreaction,PCR)检测AQP5和MUC5AC的mRNA水平,Western杂交、酶联免疫吸附试验(enzyme-linked immunosorbent as-say,ELISA)法检测AQP5和MUC5AC蛋白的变化。结果:给予SPC-A1细胞LPS 0μg/mL、5μg/mL、10μg/mL、20μg/mL、40μg/mL浓度刺激后,AQP5mRNA和AQP5蛋白表达呈LPS浓度依赖性降低,而MUC5AC mRNA和MUC5AC蛋白表达呈LPS浓度依赖性升高。LPS干预0、12、24、48h后,AQP5mRNA和AQP5蛋白呈时间依赖性降低,MUC5AC mRNA和MUC5AC蛋白升高,两者呈负相关。结论:LPS刺激后AQP5和MUC5AC的变化呈负相关。
Objective:To investigate the changes of aquaporin-5(AQPS) and mucin 5AC(MUC5ACI in SPC-A1 cell line stimu- lated by lipopolysaccharide (LPS). Methods.. After SPC-A1 cell line was stimulated by LPS, the mRNA levels of AQP5 and MUC5AC were detected by real-time polymerase chain reaction (PCR), and AQP5 and MUC5AC protein were measured by Western blot and enzyme-linked immunosorbent assay (ELISA). Results: The protein expression and transcription of AQP5 decreased while the MUC5AC expression increased in a close (0μg/mL,5μtg/mL,10 μg/mL,20μg/mL and 40 μg/mL, respectively) and time-dependent manner by LPS stimulation in SPC-A1 cell line. Conclusions: Decreased AQP5 expression was cor- related with increase of MUC5AC in SPC-A1 cell line after LPS exposure.
