枸橼酸他莫昔芬影响人卵巢癌细胞增殖机制探讨 被引量:1

摘要 目的研究枸橼酸他莫昔芬(tamoxifen citrate)对体外培养的人卵巢癌细胞SKOV-3增殖的影响及相关机制。方法采用四甲基偶氮唑盐(MTT)比色法检测0.01μmol/L枸橼酸他莫昔芬在不同的时间点对SKOV-3细胞增殖的影响;采用细胞免疫化学(immunocytochemistry)和Western blotting检测0.01μmol/L枸橼酸他莫昔芬作用于SKOV-3细胞48 h后对细胞中Bcl-2表达的影响。结果枸橼酸他莫昔芬对SKOV-3细胞的增殖具有明显的抑制作用,与对照组相比差异有统计学意义(P<0.05);枸橼酸他莫昔芬能够下调Bcl-2蛋白的表达。结论枸橼酸他莫昔芬可显著抑制SKOV-3细胞的增殖,其增殖抑制作用可能与其诱导细胞凋亡有关,而枸橼酸他莫昔芬诱导凋亡可能与其下调Bcl-2蛋白表达有关。 Objective To explore the effects of tamoxifen citrate on the proliferation of human ovarian cancer cells in vit- ro cell culture and its action mechanism. Methods MTr assay was used to determine the effects of tamoxifen citrate at 0.01 p, mol/L concentration and different time points(24,48,72 h) on the proliferation of SKOV-3 cells. The Bcl-2 expressim, in SKOV-3 cells treated with 0.01 Ixmol/L tamoxifen citrate for 48 h was analyzed by immunocytochemistry and Western blotting. Results Com- pared with the control group,tamoxifen citrate could significantly inhibit the proliferation of SKOV-3 cells in a time-dependent manners(P〈0.05 or P〈0.01 ). Tamoxifen citrate also down-regulated the expression of Bcl-2 protein. Conclusion Tamoxifen cit- rate can significantly inhibit the proliferation of SKOV-3 cells in time-dependent manners ,which may be related to the induction of apoptosis. The tamoxifen citrate-induced apoptosis might be related to the down-regulation of Bcl-2 protein.
