衰老相关基因Klotho在小鼠心脏表达的增龄性变化 被引量:2

摘要 目的通过观察昆明小鼠心脏发育过程中Klotho基因的定位及其表达。方法新生1、180、540 d昆明小鼠各5只,断颈处死,分别取出肾脏和心脏,并将心脏的右心房部分剪下;1、2、4、7、15、30、60、120、180、270、360 d和540 d昆明小鼠各10只,断颈处死后取出心脏组织;以上均提取总RNA,应用荧光定量RT-PCR技术检测Klotho mRNA的表达。结果切除右心房的心脏组织,未能检测到Klotho mRNA的表达,肾脏和右心房存在表达;昆明小鼠出生后即可在心脏检测到Klotho mRNA的表达,1、2、4、7、15、30、60、120 d小鼠相对表达量基本呈上升趋势;270、360、540 d小鼠相对表达量基本呈下降趋势。结论 Klotho基因在昆明小鼠的心脏组织中可能仅在右心房表达;120 d之内,Klotho基因在心脏组织的表达量有随增龄而增加的趋势;270 d之后,Klotho基因在心脏组织的表达量有随增龄而减弱的趋势。 Objective To examine the expression of Klotho gene during the heart development in the mouse after birth, and identify the exact region expressing Klotho gene. Methods 5 newborn mice ( Kunming strain) , 5 mice aged 180 days old, and 5 mice aged 540 days old were decapitated, the kidneys and hearts were excised, then the right atriums were removed. Expression of Klotho genes was exam- ined in mouse heart from the newborn period, through day 1, 2, 4, 7, 15, 30, 60, 120, 180, 270, 360, and day 540 respectively 10 mice; then total RNA was extracted and mRNA was amplified using real time RT-PCR. Results The transcript was not detected by the RT- PCR in mouse heart after removal the right atrium but expressed in kidney and right atrium ; Klotho mRNA was detected in the heart of newbo- rns. And the relative amounts of Klotho mRNA in the heart was upward trend, 270,360,540 day was downward trend. Conclusions In the heart of Kunming strain, Klotho gene may express solely at the right atrium. Klotho gene expression in the heart is apt to increase from 1 day to 120 day, and is apt to decrease from 270 day to 540 day.
