摘要
为探讨缺氧诱导因子-2α(HIF-2α)对人结肠癌SW480细胞凋亡的影响,本实验通过培养结肠癌SW480细胞,建立细胞缺氧模型,并通过琼脂糖凝胶电泳和流式细胞术等观察HIF-2a对人结肠癌SW480细胞凋亡的影响。结果显示:(1)单纯缺氧组细胞中HIF-2α表达显著高于常氧组、缺氧+HIF-2αsiRNA。组和缺氧+HIF-2asiRNA。组(P〈O.05);(2)单纯缺氧组与常氧组相比,细胞凋亡明显减少(P〈O.05),常氧组、缺氧+HIF-2asiRNA。组和缺氧+HIF-2asiRNA2组之间细胞凋亡数量无明显差异(P〉O.05)。结果表明,HIF-2a可抑制人结肠癌SW480细胞凋亡。
The objective of this study was to explore the influence of hypoxia inducible factor--2α(HIF-2α) on the apoptosis of SW480 cell of human colonic carcinoma. In this experiment culture colonic carcinoma SW480 cells, establishing hypoxia model and observeing the influence of HIF-2α on SW480 cells through agarose gel electrophoresis and flow cytometry were done. As results, 1)in simple hypoxia group the ex-pression of HIF-2α was higher than other three groups(i, e normal oxygen group, hypoxia+ HIF-2α siR-NA1 group and hypoxia-+- HIF-2α siRNA2 group) ( P 〈 0.05) ; 2) compared with normal oxygen group, the cell apoptosis of simple hypoxia group was significant less( P〈0.05), but among the later three groups cell apoptosis there was no significant change( P 〉0.05). Results show that HIF-2α can inhibit the apop-tosis of SW480 cell of human colonic carcinoma.
