摘要
目的观察糖心乐(TXT)对链脲佐菌素(STZ)实验性糖尿病心肌病大鼠心肌转化生长因子β_1(TGF-β_1)的影响。方法 SD大鼠腹腔注射STZ建立糖尿病模型,成模后分组,造模后八周分别给予TXT大剂量、TXT小剂量、达美康灌胃治疗,模型对照组灌生理盐水。连续四周,免疫组化法检测TGF-β_1表达情况。结果与正常对照组比较。模型组大鼠心肌组织中TGF-β_1明显升高(P<0.01);与模型组比较,TXL大剂量组和达美康组大鼠心肌组织中TGF-β_1明显降低(P<0.01);TXL大剂量组和达美康组对大鼠心肌组织中TGF-β_1的影响无明显差异(P>0.05)。结论 TXT对DM大鼠有一定的降血糖的作用,能明显改善DCMP早期心肌重构,机制可能与下调TGF-β_1在心肌中的表达,减缓心肌组织纤维化有关。
Abstract: Objective To study the effect of Tangxinle on diabetic cardiomyopathy induced by streptozotocin ( STZ), the expression of TGF - β1 in myocardium of diabetic rats were investigated. Methods SD rats were randomized into blank control group, model control group, Diamicron treatment group, Tangxinle treatment group 1 ( large dosage) and group 2 (small dosage). Animal model were established by intraperitoneal injection of STZ. After eight weeks, saline, Diamicron, large dosage and small dosage Tangxinle were given to corresponding rats for 4 weeks and the expression of TGF -β1 was examined by immunohistochemistry. Results Compared with blank control group, the level of TGF -β1 in model control group were improved obviously( P 〈 0.01 ). Compared with model control group, the levels of TGF - β1 in Tangxinle treatment group 1 and Diamicron treatment group were reduced significantly( P 〈 0.01 )but there was no significant difference between these two groups( P 〉0.05). Conclusions Tangxinle has a hypoglycemic effect and can improve the myocardial remodeling of diabetic cardiomyopathy at the early stage, the mechanism may be its decreasing the expression of TGF - β1 and alleviating the fibrosis of the myocardium.
