摘要
邻位连接技术(PLA)是近年来发展起来的一种蛋白质检测方法。该技术的核心理念来源于适体技术、连接酶技术和Taqman探针技术,其主要形式包括均相、固相和原位3种。PLA具有特异、灵敏、快速、高通量和多功能等优点,不但能够检测蛋白质、亚细胞结构和细胞等生物样本,还能检测大分子间的相互作用,现已逐步被应用于医学和生物学等众多领域。
Proximity ligation assay(PLA) is a newly developed technology for protein detection, which originated from aptamer technology, ligation chain reaction and Taqman probe real-time PCR. There are mainly three types of PLA, namely homogenous PLA, solid-phase PLA and in situ PLA. This technology is specific, sensitive, rapid, high-throughput and multiftmctional. PLA can be used for detection of biospecimens and analysis of interaction between macromolecules, so it has been extensively applied in many fields, such as medicine and biology.
