摘要
目的:研究β-catenin蛋白及DKK-1(Dickkopf-1)蛋白在宫颈鳞癌中的表达,探讨两者在宫颈鳞癌侵袭转移中的作用和机制,分析DKK-1表达水平改变时对宫颈鳞癌的侵袭能力和Wnt信号通路活性的影响。方法:应用免疫组织化学染色方法检测54例宫颈鳞癌组织、20例正常宫颈组织中β-catenin的异常表达和DKK-1蛋白的阳性表达,并分析不同宫颈组织临床及病理因素之间的关系。结果:宫颈鳞癌中DKK1蛋白的阳性表达低于正常宫颈组织,而β-catenin蛋白的异常表达高于正常宫颈组织(P<0.01);DKK-1和β-catenin蛋白在宫颈鳞癌中的表达与肿瘤分化程度、临床分期及有无淋巴转移有关(P<0.05)。宫颈鳞癌中DKK1的阳性表达与β-catenin的异常表达呈负相关(r=-0.465,P<0.01)。结论:DKK-1表达的减少或缺失与宫颈鳞癌的发生和发展密切相关,其机制可能与活化Wnt/β-catenin信号通路有关。
Objective: To study the expressions of ~ - catenin protein and DKK - 1 protein in cervical squamous cell carcinoma, explore their roles in invasion and metastasis of cervical squamous cell carcinoma and the mechanism, analyze the effect of change of expres- sion level of DKK - 1 on invasive ability of cervical squamous cell carcinoma and activity of Wnt signal pathway. Methods: Immunohisto- chemical staining was used to detect the abnormal expression of ~ - catenin protein and positive expression of DKK - 1 protein in 54 speci- mens of cervical squamous cell carcinoma tissue and 20 specimens of normal cervical tissue, the relationships between clinical factors and pathological factors in different cervical tissues were analyzed. Results : The positive expression rate of DKK - 1 protein in cervical squamous cell carcinoma was significantly lower than that in normal cervical tissue, while the abnormal expression rate of ~ - catenin protein in cervical squamous cell carcinoma was significantly higher than that in normal cervical tissue (P 〈 0. 01 ) . There was a correlation between the ex- pressions of DKK - 1 protein and ~ - catenin protein in cervical squamous cell carcinoma and degree of differentiation, clinical staging, and lymph node metastasis or not'( P 〈 0. 05 ) . There was a negative correlation between positive expression of DKK- 1 protein and abnormal expression of ~ -catenin protein in cervical squamous cell carcinoma (T = -0. 465, P 〈 0.01 ) . Conclusion: The decrease or deletion of DKK - 1 protein expression is closely related to occurrence and development of cervical squamous cell carcinoma, the mechanism may be cor- related with activated Wnt/β -catenin signal pathway.
