摘要 研究建立了反相高效液相色谱-荧光检测法(HPLC-FLD)测定玉米低聚肽中丙氨酸-酪氨酸(AY)的方法。色谱柱选为Agilent Hypersil ODS柱(125 mm×4.6 mm×5μm);荧光检测激发波长为340 nm,发射波长为450 nm;洗脱程序为梯度洗脱,流速为1.0 mL/min。玉米低聚肽样品经邻苯二甲醛(OPA)柱前自动衍生,能使AY与样品中游离氨基酸、小肽等在30 min内得到良好分离,并且AY浓度在0.1~1 000.0 mg/L时其浓度与相应峰面积有良好的线性关系,最低检测限(以信噪比为3计)为0.005 mg/L。样品加标各AY的回收率为97.8%。该法快速准确,简便灵敏,分离度高,能够满足玉米低聚肽中AY的检测要求。 A high-performance liquid chromatographic(HPLC) procedure for determining AY was described.The method was based on a reversed-phase column(Agilent Hypersil ODS column,125 mm×4.6 mm×5 μm) with precolumn derivatization withο-phthaldialdehyde and fluorometric detection(excitation wave length was 340 nm and emission wave length was 450 nm).AY were separated using gradient solution with the flow rate 1.0 mL /min.It allowed simultaneous determination of AY,free amino acids and other peptides in 30 min.Linearity for each amine was observed from 0.1 to 1 000.0 mg/L.Detection limit was 0.005 mg/L.The average recovery of AY from corn oligopeptides was 97.8%.The method was simple,fast,and reliable.It could be used to detect AY in corn oligopeptides.
