摘要
目的寻找类风湿关节炎的疾病相关的新的自身抗原。方法从已知的类风湿关节炎滑膜组织中高表达的蛋白分子中,通过淋巴细胞增殖试验及细胞因子检测,筛选出类风湿关节炎特异性自身抗原,并分析血清中抗原水平与疾病活动度的相关性。结果磷酸丙糖异构酶能特异性刺激RA外周血淋巴细胞增殖并分泌INF-γ、TNF-α及IL-17等细胞因子;RA血清中磷酸丙糖异构酶含量明显高于正常人,且血清磷酸丙糖异构酶水平与疾病活动度评分呈明显正相关。结论磷酸丙糖异构酶是类风湿关节炎新的疾病相关性自身抗原。
Objective To find a novel disease associated autoantigen for rheumatoid arthritis.Methods From the known high expression proteins in rheumatoid arthritis synovial tissue,through lymphocytes proliferation assay and cytokine detection,we screened out the specific autoantigen for rheumatoid arthritis and analyzed the correlation between serum antigen levels and disease activity.Results Triosephosphate isomerase stimulated RA peripheral blood lymphocyte proliferation specifically and secretion of INF-γ,TNF-alpha and IL-17 cytokines.The serum levels of triosephosphate isomerase from rheumatoid arthritis were significantly higher than normal serum,and the serum levels of triosephosphate isomerase were positively correlated with the disease activity score.Conclusion Triosephosphate isomerase is a novel disease associated autoantigen for rheumatoid arthritis.
