金属硫蛋白在乳化异氟醚减轻小鼠肝脏缺血再灌注损伤中的作用 被引量:1

摘要 目的探讨乳化异氟醚预处理对小鼠肝脏缺血再灌注损伤(HIRI)的保护作用,并证明金属硫蛋白参与这一作用。方法将小鼠制成HIRI模型,野生型小鼠30只,随机分为生理盐水组(S+组)30%脂肪乳剂对照组(I+组)、8%乳化异氟醚预处理组(E+组),每组10只;将金属硫蛋白基因敲除小鼠(MT--)10只,8%乳化异氟醚预处理组(E-组)。测定肝脏缺血30min再灌注1h后血清ALT、AST、MDA、SOD、肝组织病理学改变、Tunnel染色及免疫组织化学金属硫蛋白(MT)的结果。结果①野生型小鼠中:与S+、I+组相比,E+组肝脏缺血再灌注后血清ALT和AST含量显著降低,肝细胞匀浆MDA含量减少,SOD含量增加,肝组织病理学损害明显减轻,肝细胞凋亡率下降,MT免疫组化显示较高(P<0.05);②野生型与基因敲除小鼠比较:与E+组相比,E-组的ALT、AST。MDA含量升高,MDA含量下降,肝细胞凋亡率上升,MT免疫组化显著下降(呈阴性)(P<0.05)。结论乳化异氟醚预处理对HIRI有保护作用。金属硫蛋白被乳化异氟醚诱导而参与这一作用。
