摘要
目的建立通络解毒凝胶剂中士的宁和盐酸小檗碱的反相高效液相分析法。方法采用反相高效液相色谱法测定含量,色谱柱为C18(250 mm×4.6 mm,5μm),柱温为30℃,流动相为甲醇∶乙腈∶25 mmol/L磷酸二氢钠溶液(30∶30∶40),流量为0.9 mL/min,检测波长为254 nm。结果盐酸小檗碱在0.096~0.576μg范围内与峰面积线性关系良好,回归方程为A=3330.1577C-11.0256,r=0.9998,平均回收率为101.5%,RSD=2.7%。士的宁在0.056~0.336μg范围内与峰面积线性关系良好,回归方程为A=2528.7317C-9.8933,r=0.9996,平均回收率为97.6%,RSD=2.5%。结论该方法灵敏、准确,重现性好,可作为通络解毒凝胶剂中士的宁和盐酸小檗碱的质量控制标准。
Objective:To establish a method for identification of strychnine and berberine in Tongluojiedu Gel Agent. Methods:The determination was performed by RP-HPLC using C18 column(250×4.6 mm,5 μm). The col- umn temperature was 30 ℃. The mobile phase consisted of MeOH:ACN:25 mmol/L NaH2PO4 (30:30i40). The UV detection wavelength was 254 nm and the flow rate was 0.9 mL/min. Results:The calibration curve of berberine hydro chloride was in good linearity over the range of 0.096-0.576 μg. The regression equation was A = 3330.1577C-11.0256,r =0.9998. The mean recovery was 101.5% (RSD=2.7%). The calibration curve of strych- nine hydro chloride was in good linearity over the range of 0.056-0.336 μg. The regression equation was A = 2528.7317C-9.8933,r =0.9996. The mean recovery was 97.6% (RSD=2.5% ). Conclusion:The method is sensi- tive,accurate and reproducible.
