目的:探讨转录因子Slug基因沉默对前列腺癌细胞LNCaP缺氧后上皮间质转化(Epithelial-mes-enchymal transition,EMT)过程的影响及其相关的分子机理。方法:利用shRNA转染技术建立Slug沉默LNCaP细胞系,通过缺氧诱导EMT。采用western blot检测EMT发生的标志分子E-cadherin和N-cadherin蛋白的表达水平变化。Transwell侵袭实验评价细胞侵袭能力的改变。结果:成功建立Slug-shRNA稳定转染细胞系;缺氧诱导后,对照组E-cadherin表达水平明显降低,而N-cadherin表达水平明显升高,但是,在Slug沉默组,二者表达无明显变化;Slug沉默抑制了缺氧诱导的细胞侵袭能力的增加。结论:Slug基因沉默能够抑制缺氧诱导前列腺癌细胞EMT的发生;Slug与前列腺癌转移密切相关。
Objective: To study the effects of Slug silencing on the hypoxia-induced epithelial-mesenchymal transition (EMT) of LNCaP prostate cancer cells and its underlying mechanism. Method:We constructed a Slug-si- lencing LNCaP cell line by stable transfection of Slug shRNA. After exposure to hypoxia, two EMT markers, E- cadherin and N-cadherin were determined by western blot. We also detected cell invasion in LNCaP cells after hy- poxia. Result:We successfully constructed a Slug silencing cell line. Hypoxia induced decreased expression of E- cadherin and increased N-cadherin in control cells. The changes of E-cadherin and N-cadherin were inhibited by Slug silencing. Slug silencing also inhibited the invasion ability of LNCaP cells under hypoxia. Conclusion:Slug si- lencing can inhibit the hypoxia-induced EMT in LNCaP cells. Slug might be involved in prostate cancer metasta- sis.
