摘要 目的:建立测定蛋白质粉中L-半胱氨酸含量的方法。方法:采用高效液相色谱法。色谱柱为AgilentZORBAXSB-C18,流动相为0.1%磷酸(pH=2.05)-乙腈(80∶20),检测波长为210nm,流速为0.6mL·min-1,柱温为室温。结果:L-半胱氨酸检测浓度线性范围为0.02~1.0mg·mL-(1r=0.9990),平均回收率为98.76%,RSD=1.82%,检测限为0.01mg·mL-(1S/N≥3)。结论:本方法操作简便、结果准确可靠、重复性好,可用于蛋白质粉中L-半胱氨酸的含量测定。 OBJECTIVE: To develop a method for the content determination of L-cysteine in the protein powder. METHODS: HPLC method was adopted. The determination was performed on Agilent ZORBAX SB-C18 column with mobile phase consisted of 0.1% phosphoric acid (pH=2.05)-acetonetrile (80:20) at the flow rate of 0.6 mg·mL^-1. The detection wavelength was 210 nm and the column temperature was set at room temperature. RESULTS: The linear range of L-cysteine was 0.02-1.0 mg·mL^-1(r= 0.999 0) with an average recovery of 98.76% (RSD=l.82%). The detection limit was 0.01 mg·mL^-1(S/N≥3). CONCLUSION: The method is simple, accurate, reliable and reproducible, and it is applicable for content determination of L-cysteine in the pro-tein nowder.
