摘要
目的:探讨丹参对大鼠肢体缺血再灌注(LIR)后多器官水肿的预防作用。方法:Wistar大鼠24只随机分为对照组(C组)、缺血再灌注组(I/R组)和丹参预处理组(SM组)。用橡皮带结扎动物双后肢根部4h,然后松解橡皮带恢复血流灌注4h制作大鼠肢体缺血再灌注模型,SM组在再灌注前30min经尾静脉注射丹参注射液5mL/kg。手术取血处死动物,准确留取每只动物的心、肝、肾、肺、脑、肠及骨骼肌组织各1g。恒温烘干后称其干重并计算各标本的湿干重比值(W/D)。采用生物化学方法测定血浆超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)活性及丙二醛(MDA)水平。光镜下观察各组织的形态学变化。结果:LIR后各组织W/D均增加(P<0.05),血浆SOD活性降低,而XOD活性和MDA水平增加(P<0.05),镜下可见各组织出现结构紊乱、组织间隙增宽和炎细胞浸润等病理改变。而SM组与I/R组比较,血浆SOD活性回升,而XOD活性和MDA水平降低(P<0.05),镜下组织病理学变化有所减轻。结论:LIR可导致多器官水肿,氧化应激是其重要机制之一。丹参可通过抗氧化发挥抗组织水肿的作用。
Objective: To investigate the preventive role of salvia miltiorrhiza on multiple organ edema followed by hind limb ischemia-reperfusion (LIR) in rats. Methods: Twenty-four Wistar rats were randomly divided into 3 groups, control group (group C), ischemia-reperfusion group (group I/R) and salvia mihiorrhiza group (group SM). The model rats were made by 4 hours ischemia and 4 hours reperfusion in hind limbs. Salvia miltiorrhiza (5 mL/kg) was injected by tail vein in group SM 30 minutes before reperfusion. Animals were killed to take blood samples and take 1 gram tissues of heart, liver, kidney, lung, brain, intestine and skeletal muscle from every animal respectively. Specimens were weighed after baking (60 ~C, 55 hours). The ratio of wet and dry (W/D) was calculated. The plasma levels of superoxide dismutase (SOD), xanthine oxidase (XOD) and malondialdehyde (MDA) were detected by biochemical methods. The morphologie changes of those tissue samples were ob- served with microscope. Results: It was found that after suffering from ischemia-reperfusion, the ratio of W/D in all Speci- mens increased (P 〈 0.05). The plasma values of SOD decreased but XOD and MDA increased obviously (P 〈 0.05). After LIR, structure disorder, broaden interstitial and infiltration of inflammatory cells were observed in tissues of samples. Com- pared with I/R group, the level of SOD increased and values of XOD and MDA decreased in SM group(P 〈 0.05). Pathologi- cal changes were reduced in all samples of tissues. Conclusion: Limb ischemia-reperfusion in rats can lead to multiple or- gan edemas, in which oxidative stress is one of the important mechanisms. Salvia miltiorrhiza might play the role of anti-ede- ma by anti-oxidation.
