NPY增强剂和拮抗剂对大鼠垂体腺瘤影响的实验研究

摘要 目的研究神经肽Y(NPY)在大鼠垂体腺瘤中的作用。方法雌鼠48只分为:A(雌激素)组雌二醇0.5mg/次;B(NPY增强剂)组雌二醇0.5mg/次和增强剂0.6mg/次;C(NPY拮抗剂)组雌二醇0.5mg/次和拮抗剂0.06mg/次;D(对照)组芝麻油0.5ml/次;以上均隔日腹腔注射/次,12周后处死。取血和垂体进行病理,免疫组化和放免检测血浆NPY和血清PRL浓度。结果(1)A和B组血浆NPY及PRL浓度均比D组增高,P〈0.05。(2)免疫组化NPY:D组垂体瘤(+++),A和B组(+~++),C组仍较强(++~+++);PRL:A和B组均(+++)。C组较D组强。结论NPY在大鼠垂体腺瘤的发生、发展中起了非常重要的调节作用。 Objective To study the action of NPY on thepathogenesis and development of pituitary adenoma of rat. Methods 48 female 4 - week - old Wistar rats were divided randomly into A group (estrogen) ,which the rats were injected with estradiol benzoate 0. 5 mg/each time. B group ( NPY enhancer and estrogen) ,which the rats were injected with estradiol benzoate 0. 5 rag/each time and enhancer of NPY 0. 6 rag/each time. C group (NPY antagonist and estrogen), which the rats were injected with estradiol benzoate 0. 5 mg/each time and antagonist of NPY 0. 06 rag/each time. D group(control) , which rats were injected with sesame oil 0. 5 ml/each time. Above injection time was every other day for 12 weeks and injection methode was peritoneal injection for each rat. Rats of every groups were killed after model of pituitary adenoma established for the first, second and third month respectivly. At the same time, blood serum PRL and plasma NPY concentrations were measured with radioimmunoassay (RIA) kit, and the pathological and immunohistochemical analysis were performed on the surgically removed pituitary adenomas. Results 1. ( 1 ) The concentration of PRL in blood serum in A and B groups was higher than D group,P 〈0. 001. (2) The plasma NPY concentration in A and B groups was higher than D group, P 〈0. 05 and P 〈 0. 02. 2. Immunohistochemical analysis : ( 1 ) NPY : the expression of pituitary tissues was the highest ( + + + )in D group, high of pituitary adenoma ( + - + + ) in A and B groups, and higher of pituitary adenoma ( + + - + + + ) in C group. (2) PRL:the expression of pituitary adenoma was the highest ( + + + ) in A and B groups, higher at C group than D group. 3. Pathology: indicating that the pathologic change between C and Dgroups was similar nearly. There were disappearance of normal pituitary histological structure in A and B groups. Pituitary cells were heterotypical obviously. Conclusions NPY may play a critical regulation role in thepathogenesis and development of pituitary adenoma, as well as endocrine changes in rat.
