摘要
目的:建立竹叶柴胡中黄酮类成分(芦丁、槲皮素、山奈素和异鼠李素)HPLC的含量测定方法,为该药材的开发利用提供科学依据。方法:采用C18柱,流动相为甲醇(A)-0.1%磷酸(B)梯度洗脱,流速0.8 mL·min-1,检测波长360 nm,柱温35℃,进样体积10μL。结果:芦丁在667.8~33 390 ng(r=0.999 8),槲皮素在216.0~10 798.4 ng(r=0.999 9),山奈素在27.5~1 372.8 ng(r=0.999 9),异鼠李素在130.6~6 528.0 ng(r=0.999 8)线性关系良好,平均加样回收率分别为100.3%(RSD 0.78%),94.5%(RSD 1.13%),94.5%(RSD 1.15%),94.1%(RSD 1.09%)。结论:该法在同一色谱系统中同时测定芦丁、槲皮素、山奈素和异鼠李素的含量,操作简便、准确性高、重复性好,用于竹叶柴胡药材的质量评价。
Objective:To develop a method by HPLC for determination of flavonoid of Bupleurum marginatum.Method: Agilent Zorbax SB C18 column(4.60 mm×150 mm,5 μm),mobile phase methanol-0.1% phosphoric gradient elution,flow rate 0.8 min·mL-1,detected wavelength 360 nm.Result:The linear ranges of rutin,quercetin,kaempferol and isorhamnetin were 667.8-33 390 ng(r=0.999 8),216.0-10 798.4 ng(r=0.999 9),27.5-1 372.8 ng(r=0.999 9),and 130.6-6 528.0 ng(r=0.999 8),the average recovery rates were 100.3%(RSD 0.78%),94.5%(RSD 1.13%),94.5%(RSD 1.15%) and 94.1%(RSD 1.09%).Conclusion:The HPLC method is simply and rapid,and suitable to assess qualitative of B.marginatum.
