摘要
目的:应用16SrRNA测序及序列分析方法来鉴定一株来自实验室饲养小鼠的细菌。方法:对可疑小鼠的血液进行细菌培养,革兰氏染色观察细菌菌株的表型,PCR扩增菌株16SrRNA并进行测序,将测序序列与GenBank数据库的菌株序列进行进化树和基因距离分析。结果:本研究鉴定的菌株为革兰氏阳性表皮葡萄球菌。结论:16SrRNA基因测序鉴定细菌的方法适用于非专业研究人员;实验动物也可能含有病原体,进行动物实验时要有防护措施。
Objective: To identify a bacterial strain isolated from a laboratory breeding mouse by 16S rRNA gene sequencing.Methods: Suspected bacteria strain was isolated from a laboratory-breeding mouse.Gram stain was carried out to observe the bacterial phenotype.The bacterial 16S rRNA was amplified by PCR and sequenced.The resultant sequence was used to perform phylogenetic and alignment analysis by comparing with the known sequences in the GenBank.Results: The bacterial strain isolated in this study was proved to be Staphylococcus epidermidis.Conclusion: 16S rRNA gene sequencing was suitable for researchers to identifiy unkown bacterial strains.Laboratory animals are potential sources of pathogens and correct measures should be taken to protect infection by animal pathogens.
