舒脉胶囊对血管紧张素Ⅱ诱导的血管平滑肌细胞迁移的影响 被引量:1

摘要 目的观察舒脉胶囊及其拆方药物血清对血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)诱导的血管平滑肌细胞(vascular smooth muscle cells,VSMC)迁移的影响。方法采用组织贴块法进行VSMC培养,AngⅡ10-7 mol/L作为刺激因子,将药物血清分为舒脉胶囊全方组、活血化瘀拆方组和补脾益肾拆方组,测微尺测量细胞迁移的距离,免疫组织化学法测量基质金属蛋白酶-2(metalloproteinase-2,MMP-2)的表达情况。结果与AngⅡ组比较,舒脉胶囊全方组、活血化瘀拆方组细胞迁移距离显著减小(P<0.01),MMP-2表达减少(P<0.01),其中舒脉胶囊全方组细胞迁移距离显著小于活血化瘀拆方组(P<0.01)。结论舒脉胶囊通过抑制MMP-2的表达而抑制VSMC迁移,全方作用优于拆方。 Objective To observe the effects of Shumai Capsules (a traditional Chinese medicine recipe for invig- orating spleen- and kidney-qi and activating blood circulation) and its component recipes on angiotensin II (AngII) induced migration of vascular smooth muscle ceils (VSMCs). Methods VSMCs were cultured using tissue ex- plants method. Ang Ⅱ 10-Tmol/L was used as a stimulating factor. The medicinal sera were divided into overall Shumai Capsules group, activating blood circulation group,and invigorating spleen- and kidney-qi group. The mi- gration distance was measured using micrometer. The expression of metalloproteinase-2 (MMP-2) was assayed u sing immunohistochemistry. Results Compared with those in the AngH group, the migration distance of VSMCs and the expression of MMP-2 were significantly decreased in the overall Shumai Capsules group and activating blood circulation group (P〈0.01). The migration distance of VSMCs in the overall Shumai Capsules group was significantly lower than that in the activating blood circulation group (P〈0. 01). Conclusion Shumai Capsules can inhibit the migration of VSMCs by downregulating the expression of MMP-2. The effect of its overall recipe is better than that of its component recipes.
