摘要
以5-氨基水杨酸为模型药物,以醋酸纤维素为衣膜材料,采用浇膜法制备游离膜,分别考察3种致孔剂(壳聚糖、高脂果胶和果胶钙)制备的游离膜其透过性、膨胀性及结肠降解性,以筛选出具有良好结肠释药性的致孔剂。研究表明,以果胶钙为致孔剂的游离膜在胃和小肠的通透性最小,同时具有优良的结肠降解特性,果胶钙较适合作为结肠定位制剂的致孔剂。
This study was designed to screen the excellent pore-forming agent in colon-specific membrane, and 5aminosalieylic acid was selected as the model drug. Free membranes were prepared by film casting, among which cellulose acetate were used as the coating membrane material, ehitosan, high ester pectin and calcium pectate were used as the pore forming agent, respectively. The permeability, swelling properties and colon-specific degradability of the free membrane were examined. The free membrane using calcium pectate as pore-forming agent owned the smallest permeability in gastric juice and small intestine fluid, and exhibited significant colonspecific degradability. Calcium pectate can be a good pore-forming agent for colon-specific preparation.
