蜀葵花总黄酮正交提取工艺研究

Study on Extraction Process of Total Flavones in Althaea rosea L.

目的:研究蜀葵花总黄酮提取工艺及含量测定的最佳方法。方法:利用比色法测定蜀葵花中总黄酮含量,以总黄酮提取率为考察指标,利用正交试验设计考察乙醇体积分数、料液比、提取时间及提取温度对蜀葵花总黄酮提取工艺的影响。同时,比较不同测定方法所测得的结果。结果:蜀葵花总黄酮最佳提取工艺为:乙醇体积分数为60%、料液比为1:12、提取时间为2h、提取温度为90℃。不同测定方法测定的结果差异不大。结论:醋酸-醋酸钠缓冲溶液和AlCl3显色的方法及提取工艺简单、稳定、可行,为蜀葵花总黄酮的开发应用提供了依据。

Objective：To study the optimum extraction process and assaying method of total flavones in Althaea rosea L.Method：Colorimetric method was used to determine the total flavones of A.rosea L.Orthogonal experiment was used for the study on the effects of ethanol concentration,volume,extraction time and temperature on extraction process of total flavones from A.rosea L.Furthermore,the results of different testing methods were compared.Results：The optimum extraction process of total flavones from A.rosea L.was 1：12 solid-liquid ratio,60% ethanol and 2h for extraction time under 90℃.There was no significant difference between the different testing methods.Conclusion：The colorimetric method of HAc-NaAc and AlCl3 and extraction process are simple,stable and efficient which offer basis for the effective use of A.rosea L.
