摘要
采用柠檬酸提取法,在单因素实验的基础上,确定柠檬酸溶液的提取pH,并选取提取时间、提取温度和液料比为自变量,以海带多糖得率和DPPH自由基清除能力IC50值为响应指标,利用响应面分析方法,确定柠檬酸提取海带多糖的最佳条件为:pH=2柠檬酸溶剂,提取温度120℃,提取时间4h,液料比为35:1(mL/g)。在此条件下,海带多糖得率为13.19%±0.06%,DPPH自由基清除能力IC50值为(0.82±0.011)mg/mL。在最优条件下提取的海带多糖与传统直接水提法相比,具有较好抗氧化能力,氧自由基清除能力ORAC值为(392.482±3.218)μmolTrolox/g。
This study used citric acid extraction method based on the single factor experiment,which determined the appropriate pH and selected extraction time,temperature,and ratio of solvent /material as the independent variables and then adopted response surface methodology by picking the yield of polysaccharide and its DPPH scavenging activity IC50 value as the response indicators to determine the optimum conditions of citric acid extracted polysaccharide from laminaria japonica.The optimum conditions observed were as follows:citric acid solvent of pH2,extracting temperature 120℃,extracting time 4h,and ratio of solvent/material 35:1(mL/g).Under such conditions,the yield of polysaccharide was 13.19%±0.06%,and the DPPH scavenging activity IC50 value was(0.82±0.011)mg/mL.In comparison with traditional water extraction,citric acid extraction showed good antioxidant ability and the oxygen radical scavenging capacity ORAC value was(392.482±3.218)μmol Trolox/g under the optimum conditions.
