小鼠卵巢自体及异体移植前的尿促性素体外干预对卵泡存活的影响 被引量:2

摘要 目的探讨卵巢移植前采用尿促性素(menotrophin)体外干预对小鼠半卵巢移植后卵泡存活的影响。方法 ICR雌性小白鼠随机分为5组:(1)新鲜卵巢组织自体移植组(新鲜自体移植组),卵巢摘除后直接移植至自体肾被膜下;(2)自体移植干预组,卵巢摘除后先在含尿促性素液体中体外培养3h,之后移植至自体肾被膜下;(3)自体移植无干预组,卵巢摘除后在不含尿促性素的液体中体外培养3h,之后移植至自体肾被膜下;(4)异体移植干预组,卵巢摘除后在含尿促性素的液体中体外培养3h,之后移植至异体肾被膜下;(5)去势对照组。术后72h观察移植物存活率与移植物内正常卵泡计数,术后1个月观察各组动物的动情周期恢复情况、移植物的发育情况,测定血清卵泡刺激素(follicle-stimulating hormone,FSH)和雌二醇水平。结果移植后72h,新鲜自体移植组和自体移植干预组存活卵泡数明显高于自体移植无干预组与异体移植干预组(均为P<0.01)。新鲜自体移植组、自体移植干预组和自体移植无干预组动物的动情周期恢复率均为100%,明显高于异体移植干预组的67%(均为P<0.05)。动情周期恢复所需时间以新鲜自体移植组和自体移植干预组最短。自体移植干预组的卵泡数多于自体移植无干预组,但均低于新鲜自体移植组。与去势对照组比较,其他4组的FSH值均较低(均为P<0.01),雌二醇值均较高(均为P<0.01),其中新鲜自体移植组和自体移植干预组的雌二醇水平高于自体移植无干预组和异体移植干预组(均为P<0.01)。结论对移植卵巢给予尿促性素体外培养可提高移植后的卵泡存活率,是提高卵巢移植效果的有效途径。 Objective To investigate the effects of menotrophiu in-vitro intervention on survival grafts ovarian follicles in mouse half ovary transplantation. Methods ICR mice were randomly divided into five groups : ( 1 ) fresh ovarian tissue autotransplautation group (FFG). Ovaries were autotransplanted under the renal capsule after excised. (2) autotransplantation with intervention group ( AUHIG). Ovaries were auto- transplanted under the renal capsule after cultured for 3 hours in the medium contain with menotrophin. ( 3 ) autotransplantation without intervention group (AUHNG). Ovaries were autotransplanted under the renal cap- sule after cultured for 3 hours in the medium without menotrophin. (4) allotransplantation with intervention group (ALHIG). Ovaries were allotransplanted under the renal capsule after cultured for 3 hours with menotro- phin. ( 5 ) ovariectomized control group (OVXG). The survival rate and the number of normal ovarian folliclesof graft were observed at 3 days after operation. At a month after operation, the recovery of estrous cycle and the development of the graft were observed, and the levels of follicle-stimulating hormone (FSH) and estradiol were detected. Results The number of survival follicles in b^G and AUHIG was significantly higher than that of AUHNG and ALHIG at 72 h after transplantation ( all in P 〈 0.01 ). The recovery of estrous cycle were 100% in FTG, AUHIG and AUHNG, which were significantly higher than that in ALHIG with 67% (all in P 〈 0. 05 ). And the recovery time of estrous cycle was shortest in FTG and AUHIG. The number of follicles in AUHIG was more than that in AUHNG, but less than that in FTG. Compare with OVXG, the levels of FSH were lower and the levels of estradiol were higher in the other 4 groups ( all in P 〈 0.01 ). And the levels of es- tradiol in TFG and AUHIG were higher than that in AUHNG and ALHIG ( all in P 〈 0. 01 ). Conclusions The menotrophin intervention of transplant ovary in-vitro culture can increase the survival rate of transplant follicle, which is an effective way to improve the function of ovary transplantation.
