摘要
目的研究地塞米松干预哮喘大鼠模型撤停不同阶段(撤停前、撤停中、撤停后)合用淫羊藿女贞子的协同抗炎及其对激素不良反应的改善作用。方法给予哮喘大鼠模型腹腔注射地塞米松2周治疗后,逐周开始撤停地塞米松,撤停后继续观察2周,在激素应用的不同阶段配合灌胃淫羊藿女贞子,观察支气管肺泡灌洗液(BALF)IL-4、INF-γ、IL-5含量;血清皮质醇(COR)、血浆促肾上腺皮质激素(ACTH),下丘脑促肾上腺皮质激素释放激素(CRH)含量;BALF中糖皮质激素受体(GCR)蛋白表达。结果淫羊藿女贞子合用激素干预各组均有一定程度的下调哮喘模型大鼠BALF中IL-5、IL-4含量,上调IFN-γ表达,调节Th1/Th2失衡;升高血清COR含量;上调BALF中GCR的蛋白含量及结合力的作用。结论淫羊藿女贞子合用地塞米松可以增强激素的抗炎作用,并对内源性糖皮质激素作用途径起到保护作用,以淫羊藿女贞子合激素干预Ⅱ组作用趋势较佳。
Objective To research the anti-inflammatory effect and the improvement in hormone adverse reactions in the coordination of herba epimedii and fructus ligustri lucidi with dexamethasone(DXM)in asthma rat models at different withdrawal stages(before,during and after withdrawal).Methods After the peritoneal injection of DXM in asthma rat models for 2 weeks,DXM started withdrawing week by week.The observation lasted for 2 weeks after medicine withdrawal.At the different stages of hormone application,the gastric infusion with herba epimedii and fructus ligustri lucidi was combined.The contents of IL-4,INF-γ and IL-5 in bronchoalveolar lavage fluid(BALF),the contents of serum COR,plasma ACTH and hypothalamus CRH as well as GCR protein expression in BALF were observed.Results In every group with the coordination of herba epimedii and fructus ligustri lucidi with hormone intervention,IL-5 and IL-4 contents were up-regulated,the content of INF-γ was down-regulated and Th1/Th2 imbalance was adjusted in BALF to certain extent in asthma rat models.The serum COR content was increased.GCR protein expression and the binding force in BALF were up-regulated.Conclusion The coordination of herba epimedii and fructus ligustri lucidi with DXM intensifies the anti-inflammation of hormone and protects the effect approach of endogenous glucocorticoids.The effects in the II-group of the coordination of herba epimedii and fructus ligustri lucidi with hormone intervention are better in tendency specially.
