摘要
目的检测信号转导与转录活化因子5(STAT5)和促凋亡基因Bax在非小细胞肺癌(NSCLC)中的表达情况,探讨两者与NSCLC临床病理特征之间的关系及两者之间的关系。方法采用免疫组织化学S-P法检测STAT5和Bax在68例NSCLC及26例癌旁正常对照组织中的表达情况。结果①STAT5在NSCLC组织中的阳性表达明显高于肺正常组织(P<0.05);②Bax在NSCLC组织有一定表达,但与其在正常肺组织的表达比较,差异无统计学意义(P>0.05);③STAT5的阳性表达与NSCLC组织学类型有关,在腺癌中的表达明显高于鳞癌,与组织分化程度、TNM分期及淋巴结转移无关;④Bax的阳性表达与NSCLC的分化程度、淋巴结转移和TNM分期有关(P<0.05),与NSCLC的组织学类型无关(P>0.05);⑤Spearman相关分析显示STAT5和Bax在NSCLC中的阳性表达呈正相关。结论 STAT5、Bax可能在肺癌的发生、侵袭和转移中起重要的作用,抑制STAT5信号通路可望成为NSCLC的治疗靶点。
Objective To examine the expression of STAT5 and BAX in NSCLC and to study their relationship. Methods STAT5 and BAX proteins were detected in formalin-fixed paraffin-embedded tissue from 68 nonsmall cell lung cancer tissues and 26 normal lung tissues by SP immunohistochemical method. The levels of their expression and their relationship with STAT5 and BAX were analyzed. Results ①The positive rates of STAT5 and BAX in NSCLC were dramatically higher than those in the normal lung tissues (P〈0.01) ; ②There are some expression of Bax in NSCLC, but no difference between NSCLC and normal lung tissues was found. ③The expression of STATS was significantly correlated with histological types of NSCLC (P〈20.01), and was higher in adenocarcinomas than in squamous carcinomas; ④The expression of BAX was significantly correlated with histo- logical grading ,TNM stage and lymph node metastasis (P〈0.05), respectively,but was not correlated with the histological types of NSCLC (P 〉 0. 05). ⑤ There was significantly positive association between STAT5 and BAX. Conclusion The high expression of STAT5 and BAX in NSCLC plays important roles in occurrence, invasion and metastasis of NSCLC. The inhibition of STAT5 signal pathway may become a therapeutic target.
