摘要
目的了解泌尿生殖道解脲脲原体(Uu)对抗菌药物的敏感性,以指导临床合理用药;探讨解脲脲原体对氟喹诺酮类药物的耐药状况与gyrA基因突变之间的关系。方法 2011年深圳地区采用药敏试剂盒对Uu进行了药敏分析;随机抽取4株耐喹诺酮菌株,聚合酶链反应(PCR)扩增gyrA基因并测序,采用Blast比对法分析临床菌株与标准菌株的序列差异。结果在4株解脲脲原体菌株中,经测序分析发现gyrA基因1种突变形式(302位碱基C-T)。结论解脲脲原体对氟喹诺酮类药物的耐药可能是由于gyrA基因突变导致相应氨基酸改变所介导。
Objective To investigate the drug-sensitivity of ureaplasma urealyticum and to guide the rational use of antimicrobial agents. To study the status of fluoroquinolones resistance and GyrA gene mutation of U. urealyticum (Uu). Methods Both dilution method and drug susceptibility test kits were used to determine the drug sensitivity of Uu in Shenzhen area in 2011. Specific fragments of gyrA genes of randomly ,selected 4quinolone-resistant strains were amplified by PCR and sequenced. Blast program was used to analyze and compare the sequence of clinical isolates with standard strain T3. Results One alterations of GyrA in 4Uu strains were found(302C-T). Conclusion Resistance of Uu to fluroquinolones may have correlation to alterations of GyrA.
