原代培养乳鼠窦房结细胞肿胀激活氯电流的研究

摘要 目的记录并研究乳鼠窦房结细胞肿胀激活氯电流的电生理学特征。方法取培养乳鼠窦房结细胞,在不同干预条件下用全细胞膜片钳记录肿胀激活氯电流。结果等渗溶液中细胞背景电流较小且稳定,低渗溶液诱导激活肿胀激活氯电流,该电流呈现较明显的外向整流特点,具有容积敏感性,可被细胞外高渗溶液刺激所抑制。氯通道阻断剂tamoxifen显著抑制肿胀激活氯电流。结论培养乳鼠窦房结细胞存在肿胀激活氯电流。 Objective To record and study the electrophysiologic characteristics of swell-activated chloride current in sinoatrial node cells in neonatal rats. Methods The sinoatrial node cells cultured primarily were collected from neonatal rats, and swell-activated chloride current was recorded by using whole-cell patch clamp under different in- tervening conditions. Results The cell background current was smaller and stable in isotonic solution. Hypotonic so- lution induced and activated swell-activated chloride current. The current showed an obvious characteristic of outward rectification having sensitive to volume, and was inhibited by the stimulation of extracellular hypertonic solution. Tamox- ifen, a chloride channel blocking agent, inhibited significantly swell-activated chloride current. Conclusion There is swell-activated chloride current in primarily cuhured sinoatrial node cells in neonatal rats.
