Insulin-like growth factor 1 induces rat Schwann cell proliferation: an experimental study

目的 探索促进许旺细胞增殖的方法。方法 将原代大鼠许旺细胞进行体外培养 ,并在培养基中加入不同浓度的胰岛素样生长因子 1(IGF -1)。通过绘制细胞生长曲线及3 H -TdR渗入等方法 ,观察IGF -1对许旺细胞的作用。结果 IGF -1能使培养的许旺细胞增殖加快 ,提前进入平台期。许旺细胞的增殖与IGF -1的浓度在一定范围内呈现量效关系 ,同时IGF -1的作用并不导致许旺细胞的肥大。结论 IGF -1对许旺细胞增殖具有明显的促进作用。

Objective To investigate whether insulin-like growth factor 1 induces Schwann cell proliferation.Methods The primary Schwann cells of rat were incubated with IGF-1 at different concentrations. The influence of IGF-1 on Schwann cell was observed by means of growth curve and 3?H-TdR permeation. Results The Schwann cell incubated with IGF-1 at the concentration of 10 μg/L reached the stagnate phase one day earlier than the control. There was dose-dependent effect relationship between IGF-1 concentration and Schwann cell proliferation at concentration of 5～40 μg/L. Furthermore, IGF-1 stimulated hyperplasia but not hypertrophy in Schwann cells in vitro system.Conclusion IGF-1 has an obvious effect in promoting rat Schwann cell proliferation.
