摘要
以朝鲜碱茅PuP5CS基因(HQ637435)为目的基因,构建了PuP5CS基因的正义表达载体和反义表达载体,通过农杆菌介导的叶盘法转化烟草,研究了PuP5CS基因对烟草耐寒性的影响。结果显示,低温胁迫下,正义转基因烟草体内可溶性糖、脯氨酸、叶绿素a和叶绿素b的含量均显著高于野生型烟草,丙二醛含量增幅显著低于野生型烟草,提高了转基因烟草的抗寒性,而反义转基因烟草体内各项生理指标变化不明显。
To identify the responses to Chilling Stress in transplastomic tobacco with PuP5CS gene,sense and antisense plant expression vector of PuP5CS gene were constructed and introduced into tobacco via Agrobacterium tumefaciens.For the pBI121-PuP5CS+ transgenic plants,the levels of soluble suger,proline,chlorophyll a and chlorophyll b were significantly higher than wild-type tobacco,the level of malondialdehyde(MDA)was significantly lower than wild-type under the chilling stress,improved it′s cold resistance.But for the pBI121-PuP5CS-transgenic plants,the levels of them were not significant.
