摘要
[目的]探讨经静脉转染表达mIL-18的DNA质粒对Ⅰ型单纯疱疹病毒(HSV-1)急性感染小鼠的预防和治疗作用.[方法]经腹腔接种HSV-1建立小鼠感染模型,采用水流动力学方法将mIL-18基因转染Balb/c小鼠,比较观察实验组与对照组的生存率、血清中细胞因子水平、CTL和淋巴细胞增殖反应.[结果]转染pGEG.mIL-18的小鼠血清中IL-18及IFN-γ水平明显升高;转染pGEG.mIL-18小鼠组生存时间与对照组相比较差异具有统计学意义(P<0.005);转染pGEG.mIL-18小鼠脾淋巴细胞增殖反应明显增强,而对特异性P815.gB细胞的CTL水平无提高作用.[结论]用水流动力学方法转染小鼠IL-18基因对小鼠HSV-1急性感染有预防和治疗作用.
OBJECTIVE To study the prophylactic and therapeutic effects of intravascular administration of plasmid DNA expressing mIL-18 on acute herpes simplex virus type 1(HSV-1) infection in mice.METHODS HSV-1 infection mice models were established,and the pGEG.mIL-18 gene was transfected into Balb/c mice by hydrodynamics.The survival rate,serum concentration of cell factors,CTL and lymphocyte proliferation reaction were observed in experimental and control group animals.RESULTS The serum concentration of IL-18 and IFN-γ in mice transfected with pGEG.mIL-18 were increased obviously,and the survival time was more longer in mice transfected with pGEG.mIL-18 than in control group(P〈0.005).The lymphocyte proliferation reaction enhanced obviously in spleen of mice transfected with pGEG.mIL-18,but failed to elicit CTL level for specific P815.gB cells at a detectable level.CONCLUSION Intravascular immunization of IL-18 gene has an advantage in preventing and treating the acute infection with HSV-1 in mice.
