低密度脂蛋白受体基因与原发性高血压的相关性

摘要 [目的]探讨低密度脂蛋白受体(LDLR)基因Asn591 Asn多态性与延边朝鲜族自治州朝鲜族、汉族原发性高血压的相关性.[方法]采用聚合酶链-单链构象多态结合DNA测定序列方法检测280例原发性高血压患者和410例对照人群LDLR基因Asn591 Asn位点的多态性;应用酶联免疫吸附法测定血浆脂联素和胰岛素水平,应用氧化酶法检测三酰甘油及空腹血糖等生物化学指标.[结果]朝鲜族T等位基因频率在高血压组和对照组中分别为11.18%和20.48%,汉族分别为11.33%和20.77%,朝鲜族及汉族高血压组与对照组比较差异均有统计学意义(P<0.01);朝鲜族和汉族高血压组体重指数及体脂肪率明显高于对照组,而血浆脂联素水平显著低于对照组(P<0.05).[结论]LDLR基因Asn591 Asn位点的等位基因T可能是延边朝鲜族自治州朝鲜族和汉族原发性高血压发生的一个危险因子;高体重指数、体脂肪率和低血浆脂联素可能是延边朝鲜族自治州朝鲜族和汉族人群原发性高血压发生的重要危险因素. OBJECTIVE To investigate the correlation between the Asn591Asn polymorphism in low density lipoprotein receptor(LDLR) gene and essential hypertension(EH) in Korean-Chinese and Han nationality of Yanbian area.METHODS In 280 cases of EH and 410 of normotensive control individuals,the Asn591Asn polymorphism in LDLR gene was determined by PCR-SSCP and DNA sequencing,and the plasma adiponectin and insulin were measured by ELISA,and the triglyceride(TG) and fasting blood glucose(FPG) were detected by oxidase method.RESULTS Tallele frequency in EH groups and control groups were 11.18 %,20.48 % in Korean-Chinese and 11.33 %,20.77 % in Han nationality,respectively,and there were significant differences between two groups(P〈0.01).The body mass index(BMI) and body fat rate were higher in EH group than in control group,but the content of plasma adiponectin was less in EH group than in control group.CONCLUSION The allele T of Asn591Asn in LDLR is possible a risk factor for EH and the higher BMI,body fat rate and the lower plasma adiponectin are important risk factors for EH in Korean-Chinese and Han nationality of Yanbian area.
