Marc-145细胞无血清培养驯化、代谢分析及PRRSV增殖能力比较 被引量:4

摘要 采用逐步降低培养液中血清用量的方法对Marc-145细胞进行无血清培养驯化并考察其生长代谢、增殖PRRSV(猪繁殖及呼吸综合征病毒)的能力。结果表明,通过在细胞对数生长期更换低浓度血清的培养液或传代操作,Marc-145细胞可完全适应含5%新生牛血清的营养条件,经3次传代后,Marc-145细胞可营正常贴壁生长。然而在1%新生牛血清的营养条件下,初次培养于该条件的Marc-145细胞伪足不伸展,贴壁时间明显延长。经过在该1%血清浓度下10次传代后,Marc-145细胞可贴壁生长,维持较高的存活率,但贴壁较松。由此Marc-145细胞进一步驯化适应无血清悬浮培养条件,筛选获得能够在无血清条件下悬浮生长的Marc-145细胞,且生长状态良好。比较在不同血清浓度及不同培养方式下生长的Marc-145细胞增殖PRRSV的能力,无血清条件下悬浮生长的Marc-145细胞对PRRSV的增殖性能没有改变。
