内皮素和一氧化氮与老年动脉硬化闭塞症相互关系的探讨 被引量:25

摘要 目的 从分子生物学角度探讨内皮素 (ET)、一氧化氮 (NO)与动脉硬化闭塞症 (ASO)的相互关系。方法 选择 96例ASO患者和 86只模型 ,利用放射免疫、Griss、光电比色和逆转录PCR方法检测血浆ET、NO和血管一氧化氮合酶 (NOS)活性、ETmRNA和NOSmRNA表达情况。结果 ASO患者和模型均存在高ET血症和低NO血症 ,ASO病变血管存在低NOS活性、低NOSmRNA表达和高ETmRNA表达状态 ,并同ASO严重程度密切相关。结论 高ET血症、低NO血症、低NOS活性、低NOSmRNA表达和高ETmRNA表达状态同ASO的发生发展密切相关 ,ET、NO、NOS活性、NOSmRNA表达和ETmRNA表达的检测有助于ASO发病机制的探讨 。 Objective To investigate the relationship betwwen endothelins(ET),nitric oxide(NO),and atherosclerosis obliterans(ASO)from the point of view of molecular biology.Methods 96 ASO patients and 86 ASOrabbit models were selected or made.ET,NO,NOS activity,and ET-mRNA,NOS-mRNA expression were determinde by radioimmunoassy,colorimetry and RT-PCR methods in the plasma samples and ASO arterial samples.Results The ASO patients and models had high levels of plasma ET and low level of plasma NO which were related to severity of ASO,and the ETmRNA expressions of arterial samples were increased gradually,the NOSmRNA expressions and the NOS activity of arterial samples were decreased gradually with the time of ASO model making process.All changes above returned to normal gradually with effective PGEI1 treatment.Conclusion The abnormalitis of ET,NO,NOS activity,ETmRNA,NOSmRNA expressions are closely related to ASO severity and play the important role in the ASO pathogenesis.It is helpful to judge the ASO serverity and to evaluate the effectiveness of ASO treatments.
