羟丁酸钠和异丙酚对心肌细胞毒性作用的对比研究

Comparative study on cardiotoxicity of sodium r hydroxybutyrate and propofol in neonatal primary myocardial cell cultures

目的 :对比研究羟丁酸钠和异丙酚对原代培养大鼠心肌细胞的毒性作用。方法 :将经原代培养成活 4d后的大鼠心肌细胞分为 5组 ,每组 6孔。 C为对照组 ,其余 4组分别加入小剂量 (HL ,3× 10 - 3 m ol/L )与大剂量 (HH,3× 10 - 2 mol/L )羟丁酸钠组、小剂量 (PL ,3× 10 - 5 m ol/L )与大剂量 (PH,3× 10 - 4 mol/L )异丙酚组。各组均于实验开始后 8h终止反应 ,评定心肌细胞搏动功能、观察细胞形态学改变、测定心肌细胞酶及电解质变化。 结果 :与对照组相比 ,PH组搏动频率减慢 (P<0 .0 5 ) ,细胞形态学亦有相应变化 ,肌酸激酸释放量增加 (P<0 .0 5 ) ,碱性磷酸酶活性下降 (P<0 .0 5 )。而 HL ,HH及 PL组的上述指标均无明显变化。各组间电解质变化未见显著差异。 结论 :高浓度的异丙酚具有直接的心肌抑制作用 。

Objective:To investigate the profile of sodium r hydroxybutyrate and propofol cardiotoxicity. Methods:Four day old neonatal primary myocardial cell cultures were obtained from 2 to 3 day old Wistar rats which were divided into 5 groups, group C was control and group HL, HH, PL and PH were treated with sodium r hydroxybutyrate(3×10 -3 and 3×10 -2 mol/L ) and propofol (3×10 -5 and 3×10 -4 mol/L) for 8 h. The contractility, morphology, cytoplasmic enzyme (LDH, AST,CK and ALP) release content of myocardial cell and the concentration of electrolytes (k +, Na +, Cl - and Ca 2+ ) in the medium were measured 8 h following intravenous anesthetics administration.Results:In group PH the beating rates of neonatal myocardial cell cultures decreased significantly compared with group C ( P <0.05) . Release of CK in group PH were increased ( P <0.05) and ALP decreased, which was associated with the adverse effects on morphology. The results showed that group HL, HH and PL had little or no effect on all indices of toxicity evaluated. Electrolytes were not significantly different between all groups.Conclusion:The high concentrations of propofol has significant myocardial depressant action,while sodium r hydroxybutyrate and low concentrations of propofol have little or no effect on cardiotoxicity.
