Influence of estradiol and tamoxifen on cytotoxicity of lymphokine activated killer cells to lysis ovarian tumors

目的 :探讨雌二醇 (E2 )和他莫昔芬 (TAM)对淋巴因子激活的杀伤细胞 (L AK细胞 )杀伤卵巢癌细胞 3AO作用的影响。方法 :分别用 E2 ,TAM,E2 + TAM预处理 L AK细胞及 3AO细胞 12 h,然后以 5∶ 1效靶比将效、靶细胞混合作用 4h,用流式细胞术检测 L AK细胞杀伤活性 ,比较用药前后杀伤活性的变化。结果 :E2 预处理 L AK细胞及 3AO细胞后 ,其杀伤活性提高了 5 6 .6 %～ 199.2 % ;TAM处理后提高了 48.6 %～ 178.0 % ;E2 + TAM处理组与单用 TAM处理组作用相似。结论 :E2 及 TAM可显著提高 L

Objective: To investigate the effect of estradiol(E 2) and tamoxifen (TAM)on cytotoxicity of lymphokine activated killer cells(LAK cell) to lysis human ovarian tumors. Methods: Ovarian tumors cell line 3AO were used to evaluate the effects of E 2 and TAM. The cytotoxicity was measured by flow cytometry after LAK and tumor cells were pretreated with estradiol or tamoxifen. Results: cytotoxicity was improved after LAK and 3AO cells pretreated by E 2 .The increased cytotoxicicy was 56.6% 199.2%,while the improved cytotoxicity of LAK was 48.6% 178.0% after TAM pretreated. The same cytotoxicity was received when pretreated by both E 2 plus TAM and by TAM alone. Conclusion: Both E 2 and TAM can improve the cytotoxicity of LAK cells to lysis human ovarian tumors.
