摘要
从中国产蝮蛇蛇毒中分离纯化电泳纯度的类狼疮抗凝蛋白 ,研究其对动物体内的抗凝作用及机制。方法 :测定蝮蛇毒中类狼疮抗凝蛋白对新西兰白兔的活化部分凝血活酶时间、凝血酶原时间和凝血酶时间的影响 ;对小鼠凝血时间的影响 ;对大鼠静脉血栓和血小板聚集功能的影响 ,以及类狼疮抗凝蛋白的溶血性试验。结果 :类狼疮抗凝蛋白能显著地延长活化部分凝血活酶时间和凝血时间 ,明显地抑制静脉血栓的形成 ,无溶血作用。结论 :类狼疮抗凝蛋白具有体内抗凝血作用 ,作用机制可能与其干扰磷脂有关。
Objective:To study anticoagulant effect and mechanism of Lupus anticoagulant like protein(LALP),an electrophoresis purity LALP purified by ion exchange and gel filtraration chromatography from agkistrodon halys brevicaudus Methods:To measure prothrombin time (PT),activated partial thromboplastin time (APTT),thrombin time (TT);clotting time (CT);venous thrombosis and platelet aggregation in the presence of LALP in rabbit;mice;rat,respectively Results:It was showed that APTT and CT in the presence of LALP were markedly prolonged,the formation of venous thrombosis was obviously prevented without any hemolyzation Conclusion:LALP has anticoagulant effect in vivo,and the effective mechanism may interfere with phosphatide
