二烯丙基二硫下调PCNA抑制人宫颈癌移植瘤生长

摘要 目的探讨二烯丙基二硫(DADS)对宫颈癌HeLa细胞裸鼠致瘤的影响。方法裸鼠皮下注入人宫颈癌HeLa细胞建立人宫颈癌裸鼠异种移植模型,观察DADS对宫颈癌移植瘤在裸鼠体内生长情况的影响,HE染色观察DADS对移植瘤组织形态的影响,免疫细胞化学检测DADS对移植瘤组织增殖细胞核抗原(PCNA)表达的影响。结果 DADS可明显抑制人宫颈癌HeLa细胞移植瘤的生长,具有杀伤人宫颈癌裸鼠移植瘤细胞作用。PCNA蛋白在人宫颈癌移植瘤组织中高表达,DADS作用后PCNA蛋白表达明显下调。结论 DADS可明显降低人宫颈癌HeLa细胞裸鼠移植瘤生长,其机制可能与抑制肿瘤细胞增殖的PCNA蛋白表达有关。 Objective To investigate the effects of Diallyl disulfide (DADS) on xenografts in the nude mice of the human cervical cancer HeLa cells. Methods The human cervical cancer xenograft in nude mice model was established by subcutaneous injection of HeLa cells, and then the effect of DADS on cervical cancer xenograft tumor growth was evalua- ted. the effect of DADS on the morphology was observed by HE staining and the effect of DADS on PCNA expression of hu- man cervical cancer HeLa cells xenografts in nude mice was detected with SP immunohistochemistry. Results DADS significantly inhibited the growth of human cervical cancer HeLa cells xenografts and destructed HeLa cells in the human cervical cancer xenografts in nude mice. PCNA were highly expressed in human cervical cancer HeLa cells xenograft in nude mice model detected by SP Immunocytochemistry, and the expression was remarkably down-regulated after treatment with DADS. Conclusion DADS can significantly inhibit the growth of human cervical cancer HeLa cells nude mice xen- ografts,which may be associated with the down-regulation of PCNA expression.
