慢性高原病患者骨髓组织中Bcl-2、Bcl-XL蛋白表达及意义 被引量:4

摘要 目的研究慢性高原病(CMS)患者骨髓组织中抗凋亡蛋白Bcl-2、Bcl-XL蛋白表达情况,探讨其在CMS发生发展过程中的作用。方法选取门诊及住院确诊的16例CMS患者为CMS组,15例单纯陈旧性骨折患者为对照组。应用免疫组织化学SP法检测两组骨髓组织中Bcl-2、Bcl-XL蛋白的表达情况,并进行比较分析。结果 CMS组骨髓组织中Bcl-2、Bcl-XL蛋白阳性表达率分别为75.00%、81.25%,高于对照组的26.67%、33.33%(P均<0.01);CMS组Bcl-2、Bcl-XL阳性系数分别为4.25±3.11、4.43±3.15,高于对照组的0.87±0.83、0.53±0.51,差异均有统计学意义(P均<0.01)。CMS组Bcl-2、Bcl-XL蛋白表达阳性系数与血红蛋白浓度呈正相关(r分别为0.935、0.910,P均<0.05),Bcl-2和Bcl-XL蛋白阳性系数间呈显著正相关(r=0.928,P<0.01)。结论 CMS患者骨髓组织抗凋亡蛋白Bcl-2和Bcl-XL表达升高,这种变化可能参与CMS骨髓造血细胞凋亡异常机制。 Objective To study the expressions of Bcl-2 and Bcl-XL in bone marrow tissue of patients with chronic mountain sickness ( CMS), and explore their possible roles in the developing process of CMS. Methods The samples of bone marrow used in this study were obtained from 16 patients with CMS (CMS group) and 15 patients with old fracture (control group). The expressions of Bcl-2 and Bel-XL in bone marrow tissues were detected using the SP immunohisto- chemistry technique. The results were analyzed and compared between two groups. Results The positive rates of Bcl-2 and Bcl-XL proteins in patients with CMS were 75% and 81.25%, respectively, while 26.67% and 33.33% in control group, respectively. There were significant differences in the positive rates of Bcl-2 and Bel-XL between these two groups ( P 〈 0.01 ). The positive coefficients of Bel-2 and Bcl-XL proteins in CMS group were significantly higher than those in control group, respectively (4.25 + 3.11 vs 0.87-+ 0.83 for Bcl-2, and 4.43 -+ 3.15 vs 0.53 + 0.51 for Bel-XL) (P 〈 0.01 ). In the CMS group, the positive coefficients of Bcl-2 and Bcl-XL proteins were both positively related with hemoglo- bin (Hb) value (r values were 0.935 and 0.910, respectively, both P 〈0.05 ), and there were positive relationships in the positive coefficients of Bcl-2 and Bel-XL proteins ( r value was O. 928, P 〈 0.01 ). Conclusion Bcl-2 and Bcl-XL are expressed at higher levels in the CMS patients, which might be involved in the abnormal hematopoietic cells apoptosis of chronic mountain sickness.
