湖南省岳阳地区汉族人群造血干细胞捐献志愿者HLA-A*02亚型等位基因分布 被引量:2

Distribution Subtype HLA-A * 02 Allele of Hematopoietic Stem Cell Donation Volunteers in Hunan Province Yueyang Han Population

摘要 目的研究中华骨髓库湖南分库岳阳地区汉族人群造血干细胞捐献志愿者HLA-A*02亚型基因多态性分布特点。方法采用序列特异性寡核苷酸聚合酶链反应技术(PCR-SSOP)和聚合酶链反应-直接测序技术(PCR-SBT),对2010年1月~2011年12月采集的6000名湖南省岳阳地区汉族人群造血干细胞捐献志愿者进行HLA-A高分辨率分型。结果在6000例HLA-A基因分型结果中,共检出3124例A*02阳性,A*02低分辨基因频率为0.349,检出的等住基因达至010个,分别为A*02:01(O.124),A*02:02(0.001),A*02:03(0.036),A*02:05(0.001),A*02:06(0.034),A*02:07(0.106),A*02:10(0.003),A*02:11(0.001),A*02:249(0.001)和A*02:256(0.001)。其中A*02:01为优势基因,A*02:07,A*02:03和A*02:06为常见基因,该研究人群中还检出东南亚人群罕见型基因A*02。02,A*02:05,A*02:10和A*02:11及新近发现的罕见基因A*02:249和A*02:256。其基因频率分布与中华骨髓库湖北分库的基因频率分布比较差异无统计学意义,而与台湾慈济血库、日本、北爱尔兰、亚裔美国人的基因频率分布比较,差并有统计学意义。此人群A*02低、高分辨杂合度分别为82.0%和95.8%,其高分辨率纯合子呈现规律性分布。结论中华骨髓库湖南分库岳阳地区志愿者HLA-A*02亚基因频率呈高度多态性,与中华骨髓库湖北分库志愿者两个群体间A*02基因频率分布比较差异无统计学意义(χ^2=7.75,P〉0.05),而与台湾慈济血库、日本、北爱尔兰、亚裔美国人群的A*02基因频率分布比较差异有统计学意义(χ^2=48.54,340.45,655.63,484.9,P〈0.01)。 Objective To study Chinese bone marrow bank Hunan branch in Yueyang Han population of hematopoietic stem cell donation volunteers HLA-A * 02 subtype polymorphism distribution characteristics. Methods Using sequence-specific oligonucleotide polymerase chain reaction (PCR-SSOP) and polymerase chain reaction and direct sequencingtechnology (PCR-SBT) to type HLA-A of 6 000 hematopoietic stem cell donation volunteers of Han population in hunan Yueyang with high resolution from January 2010 to December 2011. Results In 6 000 cases of HLA-A genotyping results, there were 3 124 cases with positive A * 02, A * 02 low resolution gene frequency of 0, 349, the detection of alleles reached 10, respectively,A*02:01(0.124),A*02 : 02(0.001),A*02 : 03(0.036),A*02 : 05(0.001),A* 02 : 06(0.034),A*02 : 07 (0. 106),A* 02 : 10(0. 003),A* 02 : 11(0. 001),A* 02 : 249(0. 001) and A* 02 : 256(0. 001). Where A* 02 : 01 was a dominant gene,A* 02 : 07,A* 02 : 03 and A* 02 : 06 common genes. The study population was also detectable in Southeast Asian populations of rare genotype A * 02 : 02,A * 02 : 05,A* 02 : 10,A * 02 : 11 and newly discovered a rare gene A* 02 : 249 and A * 02 : 256. The distribution of gene frequencies and the Chinese bone marrow bank Hubei branch of the distribution of gene frequencies were not significantly different,and with Taiwan,Japan, Northern Ireland's banks, Asian Americans the distribution of gene frequencies, there was significant difference. The group A * 02 low, high resolution heterozygosity was 82 %, 95.8 % respectively,and its high resolution in homozygotes showed a regular distribution. Conclusion The Chinese bone marrow bank Hunan branch Yueyang volunteer subtype HLA-A * 02 gene frequencies were highly polymorphic, and Chinese bone marrow bank Huhei branch volunteers two groups A * 02 gene frequency distribution P (Table χ^2= 7. 75, P〉 0. 05), and Taiwan's blood bank, Japan, Northern Ireland, Asian American populations of A * 02gene frequency distribution and there was significant difference (χ^2=48.54,340. 45,655.63,484. 9, P〈 0.01).
