摘要
以9个常用狗牙根品种为对象,利用SRAP标记对其亲缘关系进行研究,构建了9份材料的指纹图谱。结果显示,从25对引物中最终筛选出10对引物组合,共扩增出230条清晰的条带,其中154条为多态性条带,平均多态性条带百分率为66.96%,材料间的遗传相似系数范围在0.606到0.867之间,变幅为0.261,说明9个狗牙根亲缘关系较近,较难区分;UPGMA聚类分析结果显示材料可聚为2类,T-419、小矮人与老鹰草聚为一类,其他6个品种聚为一类,这表明坪用性状表现相近的草坪草品种有聚类在一起的趋势。利用引物对em4-em6和me1-em6的扩增产物电泳图为基础构建的指纹图谱可以鉴定出9个狗牙根品种。
SRAP molecular marker was used to study the genetic relationships among nine accessions and construct the fingerprints of nine accessions of bermudagrass. According to the results of SRAP, 10 pairs of primers combinations were screened out from the 25 pairs of primers. Total 230 bands were clearly amplified with 10 pairs of primers, 154 fragments were polymorphic and the average percentage of polymorphic bands was 66.96%, The Nei's genetic similarity coefficient of the tested accessions ranged from 0. 606to 0. 867, and the luffing was 0. 261. The results suggested that it was more difficult to distinguish the accessions because their genetic relationships were near, and there was a rich genetic diversity among the nine bermudagrass tested. The accessions were divided into two groups based on UPGMA cluster genetic similarity. Moreover, Tifway, Tifdwalf and Tifeagle were clustered into one group. The rest were clustered into another group. It suggested that the turf clustering together. Nine bermudagrass varieties can be the electrophoresis map of products amplified by primer grass varieties with similar traits had the trend identified by using the fingerprints established pairs of em4-em6 and mel-em6.
