同时检测逍遥丸中芍药苷和槲皮素的含量研究

Simultaneous Determination of Paeoniflorin and Quercetin in XiaoYaoWan by HPLC

目的:优化同时测定逍遥丸中芍药苷和槲皮素含量的方法。方法:采用HPLC法和菲罗门Luna-C18柱(250mm×4.6mm,5μm);流动相:乙腈-0.1%磷酸,梯度洗脱;柱温:30℃,芍药苷、槲皮素的检测波长分别为230nm、254nm。流速:1.0mL.min-1(0～30min),0.8mL.min-1(30～60min)。结果:芍药苷和槲皮素的分离度很好,无其他杂质干扰,芍药苷在7.03～70.26μg/mL(r=0.999 7)、槲皮素在2.52～25.23μg/mL(r=0.999 7)范围内线性关系良好,平均加样回收率(n=6)分别为99.14%、98.69%,RSD分别为1.43%、1.40%。结论:该方法操作简便,结果准确,可用于逍遥丸中芍药苷与槲皮素的同时测定,为提高逍遥丸的质量标准提供了依据。

Objective：Optimization the HPLC method which Simultaneous determination of paeoniflorin and quercetin in XiaoYaoWan（XYW）.Methods：The HPLC analysis was conducted on Luna-C18 column（4.6 mm×250 mm,5 μm）,The mobile phase consisted of acetonitrile ： 0.1% phosphoric acid in a gradient elution.The flow rate was 1.0 mL/min at 0~30 min,and 0.8 mL·min-1at 30~60 min,The column temperature was 30℃,the detection wavelength was set at 230nm,254 nm,respectively.Results：The resolution of paeoniflorin and quercetin was good,with no miscellaneous peak.the linear range of paeoniflorin was 7.03~70.26 μg/mL（r=0.999 7）,the average recovery was 99.14 %,RSD=1.43 %（n=6）,and the linear range of quercetin was 2.52~25.23 μg/mL（r=0.999 7）,the average recovery was 98.69 %,RSD=1.40 %（n=6）.Conclusion：The method is simple,accurate,and can be used for the determination of paeoniflorin and quercetin in XYW.
