抗原热灭活对小鼠肺炎支原体ELISA检测的影响

The Effect of Antigen Heat-inactivation to ELISA Assay of Mice Infected with Mycoplasma pneumonia

目的优化小鼠肺炎支原体ELISA检测方法,降低非特异性反应。方法肺炎支原体热灭活处理后制备ELISA板,检测小鼠血清,与常规ELISA及分离培养方法相比较。结果优化后的ELISA方法平均本底值为0.03,敏感度为1:1280;优化ELISA方法与分离培养方法的符合率为99.27%,比常规ELISA方法高;PCR验证改进后的ELISA方法具有较好的准确性。结论优化后的方法准确性好,灵敏度高,对小鼠肺炎支原体的检测具有重要的实际意义。

To optimize ELISA assay for mice infected with Mycoplasma pneumonia（MP） for reducing non-specific reactions.ELISA plate was coated with MP antigen of heat-inactivated,then detected MP antibody from mice serum,compared with routine ELISA and IA.The optimized method have lower background noise value（0.03）,the titer of serum is 1：1280.And,the result of the optimized ELISA is consistent with PCR.The optimized method has good accurateness and high sensitivity.Thus,this work has important significance to the detection of MP in mice.
