摘要
目的优化小鼠肺炎支原体ELISA检测方法,降低非特异性反应。方法肺炎支原体热灭活处理后制备ELISA板,检测小鼠血清,与常规ELISA及分离培养方法相比较。结果优化后的ELISA方法平均本底值为0.03,敏感度为1:1280;优化ELISA方法与分离培养方法的符合率为99.27%,比常规ELISA方法高;PCR验证改进后的ELISA方法具有较好的准确性。结论优化后的方法准确性好,灵敏度高,对小鼠肺炎支原体的检测具有重要的实际意义。
To optimize ELISA assay for mice infected with Mycoplasma pneumonia(MP) for reducing non-specific reactions.ELISA plate was coated with MP antigen of heat-inactivated,then detected MP antibody from mice serum,compared with routine ELISA and IA.The optimized method have lower background noise value(0.03),the titer of serum is 1:1280.And,the result of the optimized ELISA is consistent with PCR.The optimized method has good accurateness and high sensitivity.Thus,this work has important significance to the detection of MP in mice.
