醚型菊酯类农药通用抗原的合成及鉴定

Synthesis and Identification of Generic Antigen for Ether Pyrethroid Pesticides

以2-(4-乙氧基苯基)-2-甲基丙醇和氯乙酸钠为原料,合成了醚型菊酯类农药通用半抗原HaptenⅠ,经1H-NMR及13C-NMR鉴定后,分别与牛血清蛋白(BSA)和卵清白蛋白(OVA)偶联,制得免疫原和包被原,经紫外光谱分析法计算得其偶联比分别为14∶1和35∶1,说明人工抗原合成成功。免疫Balb/c小鼠制备多克隆抗体,效价达1.28×105,用半抗原经间接竞争ELISA检测人工抗原的免疫原性,IC50和IC10值分别为0.2653和0.0012 mg/L,证明人工抗原具有较好的免疫原性。交叉反应表明此多克隆抗体具有良好的特异性。

2-（4-Ethoxyphenyl）-2-methylpropanol can react with sodium chloroacetate by nuclephilic substitution reaction.HaptenⅠ,as the product of the above reaction identified by 1H-NMR and 13C-NMR,was conjugated to bovine serum albumin（BSA） and ovalbumin（OVA） by the active ester method to get artificial immune antigen and coating antigen,respectively.The binding ratio of artificial antigens was detected by UV spectroscopy as 14 ∶ 1 and 35 ∶ 1.Balb/c mice were immunized with immunogen to prepare antiserum whose titer reached 1.28×105.The icELISA was conducted with the antiserum,and the IC50 was 0.2653 mg/L and the IC10 was 0.0012 mg/L.
