摘要
目的 探讨膀胱肿瘤多药耐药基因 (MDR- 1m RNA)的表达与临床化疗的相关性。方法 采用逆转录多聚酶链反应 (RT- PCR)技术 ,对 2 6例正常膀胱粘膜和 30例膀胱肿瘤 MDR- 1m RNA的表达进行了检测。结果 2 6例正常膀胱粘膜中 MDR- 1m RNA表达为 15 .4% ,16例初发膀胱癌 MDR- 1m RNA有表达为37.5 % ,14例复发膀胱癌的 MDR- 1m RNA表达为 78.6 % ,并对其进行了临床生物学行为的观察和比较。结论 多药耐药现象在正常膀胱粘膜中有表达 ,在部分初发膀胱癌中有先天性存在 ,RT- PCR方法检测膀胱肿瘤时具有相对敏感、特异、方便。
Objective To investigate the correlation between expression of multidrug resistance gene (MDR 1mRNA) in bladder tumor and its clinical chemotherapy.Method Using RT PCR technique,expression of multidrug resistance gene(MDR 1mRNA)in 26 specimen from normal bladder mucosa and 30 from bladder tumor were measured.Results The results showed that the expression of MDR 1mRNA in 26 cases of normal bladder mucosa is 15.4%,that in 16 cases of untreated primary tumor is 37.5% and that in 14 cases of the recurrent tumor is 78.6%.Correlation of the MDR 1mRNA expression with the clinical biological behavior was also observed.Conclusion The phenomenon of multi drug resistance exists in normal bladder mucosa,but untreated primary tumor has a positive MDR 1mRNA expression,which correlates with the degree of malignance.The RT PCR assay proved to be highly senitive,relatively quantitative and reliable for measuring the level of MDR 1mRNA expression in clinical specimens.
