丙型肝炎病毒高变区1模拟表位7肽细胞免疫应答作用的研究

摘要 目的探讨丙型肝炎病毒高变区1模拟表位7肽在体外刺激免疫小鼠脾淋巴细胞后的细胞增殖情况。方法选用经7肽皮下多点免疫和脾内直接注射两种免疫途径的免疫小鼠,无菌条件下分离脾淋巴细胞后,实验组分为刺激组与未刺激组,刺激组脾细胞再经7肽刺激,应用单溶液细胞增殖检测法体外检测7肽对免疫小鼠脾淋巴细胞在不同浓度和不同培养时间点增殖的影响。结果体外7肽刺激免疫小鼠脾淋巴细胞后,刺激组的脾淋巴细胞增殖反应与未刺激组比较存在促进作用,与对照组比较出现了明显的增殖反应。结论丙型肝炎病毒高变区1合成肽在体外对免疫小鼠脾淋巴细胞具有一定的促增殖作用。 Objective To study the effect of synthetic peptides(7-P) from HCV HVR1 on proliferation of mouse splenic lymphocytes in vitro.Methods Mice were immunized by 7-P through hypodermic injection or injected directly in the spleen before splenocytes were separated.After that,the splenocytes were stimulated again by 7-P in vitro,followed by MTS test to detect the effect of synthetic 7-P on splenocyte proliferation at different dose and incubation.Unimmunized mice were treated as negative control.Results 7-P could obviously promote the proliferation of splenocytes compared with control(without 7-P stimulation) and negative control groups(without 7-P immunization).Conclusion Synthetic peptides(7-P) derived from HCV HVR1 can promote splenocyte proliferation of 7-P-immunized mice in vitro.
