膀胱移行细胞癌P16基因5’CpG岛甲基化与肿瘤恶性程度关系分析

摘要 目的 :了解P16基因在膀胱移行细胞癌 (TCC)中甲基化改变及这一改变与肿瘤恶性程度的关系。方法 :采用PCR法检测 31例膀胱TCCs标本及 9例正常膀胱粘膜P16基因甲基化。结果 :正常膀胱粘膜无甲基化P16基因外元 1甲基化率为 2 9.0 3% ,外元 2为 35 .48% ;外元 1甲基化与肿瘤恶性程度有显著相关性 (P <0 .0 1) ,外元 2甲基化与肿瘤恶性程度无相关性 (P >0 .0 5 )。结论 :膀胱TCCP16基因存在甲基化 ,外元 1甲基化可使细胞增殖进一步失控而使肿瘤恶性程度增加 ,在P16基因失活中可能起更重要作用。 Objective:To explore the methylated alteration of P16 gene and the relationship between methylation of P16 gene and aagressive behavior of tumor in transitional cell carcinoma (TCC) of urinary bladder.Methods:PCR technique was used to detect the methylation of P16 gene in 31 specimens of TCCs and 9 control specimens of normal mucosas of the bladder.Results:Methylation of P16 gene was not observed in normal mucosas of the bladder,the methylated rate of exonl of P16 gene was 29.03%,exon 2 was 35.48%.A highly significant correlation (P<0.01) between the methylation of exon 1 of P16 gene and histologic grading as well as clinical staging were showed.In contrast,there was not a significant association P>0.5) between the methylation of exon 2 and the grading as well as staging.Conclusion:Methylation of P16 gene exists in TCC of bladder.Methylation of exon 1 can enhance the tumor growing out of control which results in tumor progression,and probably play an more important role in inactivation of P16 gene.
